Abstract:

This paper proposes an endogenous topography construction strategy based on PLGCL, using strain-induced phase transformations to spontaneously form anisotropic microgrooves on the membrane surface. This morphology guides BMSC adhesion, cytoskeleton organization, and osteogenic differentiation, while activating the focal adhesion—RhoA/ROCK—Lamin A/C—YAP—RUNX2 signaling pathway. After loading with SDF-1α, the material enhances stem cell recruitment and promotes new bone formation in a rat cranial defect model, showing the potential of endogenous topography and chemical signals to work together in regulating bone regeneration.

Research Background

Bone tissue engineering requires efficient, stable, and precise induction of stem cell osteogenesis. Chemical modifications or growth factor delivery are often limited by cost, stability, and microenvironment compatibility; micro/nano topographies can guide cells through contact, but the relationship between topological parameters and lineage fate is still unclear. Traditional external processing methods like photolithography and nanoimprinting are complex and might affect surface signaling. Therefore, there is an urgent need for controllable and scalable methods to construct endogenous surface topographies.

Main Content

This study uses PLGCL as the base material and regulates the transition of spherulites to a shish-kebab structure through uniaxial solid-state stretching, inducing molecular chain rearrangement and crystal reorganization, forming size-adjustable micro-grooves on the surface. The effects of stretching ratio on crystallization, morphology, hydrophilicity, mechanics, and degradation were systematically investigated to screen and optimize the morphology. Then, SDF-1α was loaded via immersion and freeze-drying to create a composite membrane with both topographical guidance and chemotactic recruitment, and its effects on BMSC behavior, mechanotransduction, and osteogenic differentiation were evaluated.

Results

The solid-state stretching induced PLGCL strain crystallization, forming a highly oriented shish-kebab structure, with parallel micro-grooves appearing on the surface. The groove size changed non-monotonically with the stretching ratio, and the optimized group had suitable width, depth, and edge curvature. The micro-grooves promoted BMSC proliferation, elongation, and cytoskeleton alignment, enhanced focal adhesion maturation and RhoA/ROCK signaling, induced nuclear deformation and YAP nuclear translocation, and upregulated osteogenic markers like Runx2, BMP-2, OCN, and COL-I. Loading SDF-1α further enhanced BMSC migration and recruitment. In animal experiments, the composite membrane promoted bone formation from the defect edges toward the center, and histology showed more mature bone integration and collagen deposition, along with reduced local inflammatory response.
